目的:探讨周期依赖性蛋白激酶5(Cdk5)2种激活剂p35和p39在出生后各发育阶段大鼠三叉神经脊束尾侧亚核内的表达变化。方法:采用Western印迹分析、免疫沉降和Cdk5活性分析,检测出生后各发育阶段大鼠三叉神经脊束尾侧亚核内Cdk5的活性变化和Cdk5、p39的表达水平。采用SPSS11.0软件包中的两两比较和扩展t检验进行统计学分析。结果:在新生大鼠三叉神经脊束尾侧亚核组织中,p39表达较低;出生后2~3同时,p39的表达最高;到成体时,其表达降低到与新生大鼠三叉神经脊束尾侧亚核组织中的相同水平。在出生后各发育阶段,大鼠三叉神经脊束尾侧亚核内Cdk5的表达一致,而Cdk5的活性逐渐减弱,新生大鼠三叉神经脊束尾侧亚核内的Cdk5活性(115.5 Kcpm)比成体中的Cdk5活性(19.0 Kcpm)约高6倍,各组之间均有显著差异(P<0.01)。结论:在出生后各发育阶段,大鼠三叉神经脊束尾侧亚核内p39的表达与p35表达结果在时间上有差异.Cdk5/p39和Cdk5/p35在出生后各发育阶段大鼠三叉神经脊束尾侧亚核内可能扮演不同的角色。
OBJECTIVE: To investigate the expression changes of p35 and p39, two kinds of cyclin-dependent protein kinase 5 (Cdk5) activators, in the subnucleus caudalis of the trigeminal nerve in each developmental stage after birth. Methods: Western blot analysis, immunoprecipitation and Cdk5 activity assay were used to detect the changes of Cdk5 activity and Cdk5, p39 expression in the caudate nucleus of the trigeminal nerve at each developmental stage. SPSS11.0 software package was used for comparison and extended t-test for statistical analysis. Results: The expression of p39 was low in the tail nucleus of the trigeminal nerve of the newborn rats. The expression of p39 was highest at 2 ~ 3 postnatal days. At the adult, the expression of p39 was reduced to the level of the trigeminal nerve bundle The same level in the caudal subnucleus. At each developmental stage after birth, the expression of Cdk5 in the tail nucleus of the tail trigeminal tract of rats was consistent with that of Cdk5. The Cdk5 activity (115.5 Kcpm) in the tail nucleus of the trigeminal nerve of the newborn rats Cdk5 activity in adult (19.0 Kcpm) was about 6-fold higher, with significant differences between the groups (P <0.01). CONCLUSION: The expression of p39 in the subnucleus caudalis posterior to the tail of the trigeminal nerve in rats is different in time from that of p35 expression.Cdk5 / p39 and Cdk5 / p35 express in the developmental stages of the trigeminal nerve Within the caudal subnucleus of the spinal cord may play different roles.
