目的:建立小剂量胺碘酮(AD)治疗心律失常时固相萃取-高效液相色谱血清浓度测定方法。方法:用Waters OA-SIS~(TM) HLB小柱预处理样品,70%的甲醇洗脱杂质,氯仿洗脱药物;采用ODS-C_8柱,流动相为甲醇-水-乙腈-四甲基乙二胺-冰醋酸(100:65:100:1:0.8),流速1.2 mL·min~(-1),柱温为45℃,二极管阵列检测器检测波长243 nm。结果:血清中AD在0.1~3.2mg·L~(-1)浓度范围内具有良好的线性关系,最低检测药物浓度为40ng·mL~(-1),日内与日间加样回收率分别为98.5%~101.3%与97.7%~99.5%(n=5),日内与日间HSD均低于5%(n=5)。结论:本方法简单、灵敏、准确、干扰小,适用于生物样品中AD的血药浓度监测。
Objective: To establish a method for determination of serum concentration of solid phase extraction and high performance liquid chromatography in the treatment of arrhythmia with low dose amiodarone (AD). Methods: The sample was pretreated on a Waters OA-SIS ~ (TM) HLB cartridge, eluted with 70% methanol and eluted with chloroform. The mobile phase was ODS-C_8 with methanol-water-acetonitrile- Diamine - glacial acetic acid (100: 65: 100: 1: 0.8) at a flow rate of 1.2 mL · min ~ (-1) and a column temperature of 45 ℃. The detection wavelength of the diode array detector was 243 nm. Results: Serum AD had a good linearity in the concentration range of 0.1-3.2 mg · L -1, the lowest concentration of test drug was 40 ng · mL -1. The daily and daily recoveries of AD were 98.5% -101.3% and 97.7% -99.5% (n = 5) respectively. The intra-day and inter-day HSD were less than 5% (n = 5). Conclusion: The method is simple, sensitive, accurate and small interference, suitable for the monitoring of plasma concentration of AD in biological samples.
