目的:观察苦豆子总碱(total alkaloid of Sophora alopecuroides,TASA)对S180荷瘤小鼠的抑瘤作用及其对小鼠血清肿瘤坏死因子(TNF-α)和白介素-6(IL-6)的影响。方法:建立小鼠移植性S180肉瘤模型。随机分为空白对照组、荷瘤对照组、环磷酰胺(CTX)组、苦豆子总碱高、中、低剂量(分别为80,40,20 mg.kg-1)组。连续给药10 d,观察小鼠一般情况;10 d后取血,处死小鼠,分离瘤块,计算抑瘤率;酶联免疫吸附法(ELISA)检测小鼠血清TNF-α,IL-6的水平。结果:TASA高剂量组肿瘤生长受到抑制(P<0.05);TASA各剂量组TNF-α含量与荷瘤对照组相比显著升高(P<0.05);TASA的高剂量组、中剂量组IL-6含量比荷瘤对照组显著升高(P<0.05)。结论:苦豆子总碱有抗肿瘤活性,其作用可能与提高荷瘤小鼠血清TNF-α,IL-6含量有关。
OBJECTIVE: To observe the antitumor effect of total alkaloid of Sophora alopecuroides (TASA) on S180 tumor-bearing mice and its effect on serum tumor necrosis factor (TNF-α) and interleukin-6 influences. Methods: The mouse transplanted S180 sarcoma model was established. They were randomly divided into blank control group, tumor-bearing control group, cyclophosphamide (CTX) group, Sophora alopecuroides high, medium and low doses (80,40,20 mg.kg-1) group. The mice were sacrificed 10 days later. The mice were killed and the tumors were separated. The tumor inhibition rate was calculated. The levels of TNF-α and IL-6 in serum were detected by enzyme linked immunosorbent assay (ELISA) s level. Results: The TASA high dose group inhibited the growth of tumor (P <0.05). The levels of TNF-α in TASA high dose group were significantly higher than those in the control group (P <0.05) -6 content than the tumor-bearing control group was significantly higher (P <0.05). Conclusion: The total alkaloids from Sophora alopecuroides have anti-tumor activity, which may be related to the increase of serum TNF-α and IL-6 in tumor-bearing mice.
