人体白血病细胞株K562/VCR对长春新硷(V-CR)具有抗药性。异搏停(Verapamil,一种冠状动脉扩张剂)能抑制VCR从细胞内主动逸出,使之累积于细胞内,而克服白血病细胞对VCR的抗药性。作者选择能诱导白血病细胞株K562(K562/VCR系的母细胞)红系统分化的几种化疗药物,对其诱导K562/VCR细胞红系统分化和异搏停克服K562/VCR细胞抗药性的作用做了研究。 K562细胞株是来自慢粒急变末期患者胸腔的渗出液,K562/VCR细胞株经体外方法建立。细胞培养于含10%胎牛血清和卡那霉素(100μg/ml)的RPMI1640
Human leukemia cell line K562/VCR is resistant to vincristine vinorelbine (V-CR). Verapamil (a coronary vasodilator) inhibits the active escape of VCR from the cell, allowing it to accumulate in cells and overcome the resistance of leukemia cells to VCR. The authors selected several chemotherapeutic drugs that could induce differentiation of the red system of the leukemia cell line K562 (the mother cell of the K562/VCR lineage), and their effects on inducing K562/VCR cell red system differentiation and ectopic arrest against the drug resistance of K562/VCR cells.
