目的 研究侵袭性骨肿瘤细胞凋亡及P53、P16、BcL - 2的表达的情况 ,并初步探讨其临床及生物学意义。方法 采用原位DNA末端标记法及免疫组化方法检测 6种共 12 3例侵袭性骨肿瘤中细胞凋亡及P53、P16、BcL - 2的表达情况 ,并统计其阳性反应率。结果 发现在几种骨肿瘤中P53、P16表达较普遍。BcL - 2的表达仅见于部分恶性肿瘤病例 ,良性肿瘤不表达。恶性骨肿瘤P53和BcL - 2基因蛋白表达阳性率均高于良性肿瘤 ,差异有显著的意义 (P值 <0 0 1)。恶性骨肿瘤细胞凋亡及P16基因蛋白表达阳性率均低于良性肿瘤 ,差异有显著的意义 (P值 <0 0 1)。结论 细胞凋亡及相关基因P53、P16的异常活动在骨肿瘤的发生、发展过程中可能起着相当重要的作用 ,而BcL - 2的表达可作为良、恶性骨肿瘤鉴别的一个参考指征。
Objective To study the apoptosis of invasive bone tumor cells and the expression of P53, P16, and BcL-2, and to discuss their clinical and biological significance. Methods The expression of P53, P16, and BcL-2 in apoptotic bone tumors were detected by in situ DNA end-labeling and immunohistochemistry. The positive reaction rates were statistically analyzed. Results It was found that P53 and P16 expression were common in several bone tumors. The expression of BcL-2 was only found in some cases of malignant tumors, and benign tumors did not express. The positive rate of P53 and BcL-2 gene protein expression in malignant bone tumors was higher than that of benign tumors, and the difference was significant (P < 0 01). The positive rate of apoptosis and P16 gene protein expression in malignant bone tumors was lower than that of benign tumors, and the difference was significant (P < 0 01). Conclusion The abnormal activity of apoptosis and related genes P53 and P16 may play an important role in the development and progression of bone tumors, and the expression of BcL-2 can be used as a reference indicator for the differentiation of benign and malignant bone tumors.
