目的了解胡桃楸提取物(JMME)治疗小鼠弓形虫感染的作用。方法建立弓形虫感染小鼠模型。将弓形虫感染的小鼠分成5组,分别以0.090、0.045和0.023mg/(kg.d)的JMME灌胃治疗,1次/d,疗程12d,同时设乙酰螺旋霉素(SM)治疗组和生理盐水(NS)对照组,观察各组小鼠的生存情况,弓形虫在小鼠体内的分布,计算胸腺指数和脾指数,MTT法测定淋巴细胞转化能力。结果JMME高、中、低剂量治疗均能推迟弓形虫感染小鼠的死亡时间,JMME低剂量治疗组和SM治疗组小鼠30d存活率分别为20.0%和15.0%,差异有统计学意义(P<0.05)。弓形虫在小鼠体内的分布以腹水居多,密度NS组高于各治疗组,肝、脾组织内弓形虫也以NS组居多。治疗后感染鼠体内的弓形虫未出现远端播散,除NS组外,其他各组脑组织内均未查到虫体。JMME高、中、低三剂量治疗组小鼠的胸腺指数分别为2.44±0.45、2.43±0.58和2.36±0.47,与NS组1.74±0.83比较差异有统计学意义(P<0.05);脾指数分别为4.35±0.42、4.41±0.25和4.43±0.13,与NS组3.17±0.06比较差异有统计学意义(P<0.05);淋巴细胞转化A570值分别为0.535±0.028、0.512±0.043和0.498±0.024,与NS组0.318±0.027比较差异有统计学意义(P<0.05)。结论JMME对小鼠弓形虫感染有一定的治疗作用。
Objective To understand the effect of Juglans mandshurica extract (JMME) on the treatment of Toxoplasma gondii infection in mice. Methods Toxoplasma infection mouse model was established. Mice infected with Toxoplasma gondii were divided into 5 groups and treated with JMME at 0.090, 0.045 and 0.023 mg / (kg · d), respectively, once per day for 12 days, and treated with acetylspiramycin (SM) And normal saline (NS) control group. The survival of mice in each group was observed, the distribution of Toxoplasma gondii in mice was calculated, the thymus index and spleen index were calculated, and the ability of lymphocyte transformation was determined by MTT assay. Results High, medium and low doses of JMME could delay the death of Toxoplasma gondii-infected mice. The 30-day survival rates of JMME low-dose and SM groups were 20.0% and 15.0%, respectively, with significant difference (P <0.05). The distribution of Toxoplasma gondii in mice was mostly in ascites, the density in NS group was higher than that in each treatment group, and that in Toxoplasma gondii in liver and spleen was also mostly in NS group. Toxoplasma gondii did not appear distal to the infected mice after treatment, except for the NS group, no other parasites were detected in the brain tissues of other groups. The thymus index of JMME high, middle and low dose treatment groups were 2.44 ± 0.45,2.43 ± 0.58 and 2.36 ± 0.47, respectively, which was significantly different from that of NS group (1.74 ± 0.83, P <0.05) 4.35 ± 0.42, 4.41 ± 0.25 and 4.43 ± 0.13, respectively, which was significantly different from that in NS group (3.17 ± 0.06, P <0.05). The A570 values of lymphocyte transformation were 0.535 ± 0.028, 0.512 ± 0.043 and 0.498 ± 0.024, Compared with NS group 0.318 ± 0.027, the difference was statistically significant (P <0.05). Conclusion JMME has certain therapeutic effect on Toxoplasma gondii infection in mice.
