目的 研究一种较好的微波和长春新碱的联合方法,用于白血病细胞的体外净化。方法 将微波和长春新碱分别单独以及联合作用于正常骨髓细胞,HL60,U937和K562细胞株,通过GMCFU和tCFU的测定来评价体外净化的效果。结果 微波和长春新碱对GMCFU,tCFU存活率均表现出抑制作用,抑制程度呈现出功率相关性和浓度相关性;在一定范围内,微波和长春新碱对3种细胞株tCFU存活率的抑制显著大于GMCFU(P<0.05或P<0.01);微波和长春新碱联合作用对3种细胞株的杀伤作用比单用长春新碱有显著性增加(P<0.05),而不显著增加对GM-CFU的抑制(P>0.05)。结论 微波和长春新碱联合用于白血病细胞的体外净化能产生协同效应,值得进一步研究。
Objective To study a better combination of microwave and vincristine for the in vitro purification of leukemia cells. Methods The microwave and vincristine were used separately and in combination with normal bone marrow cells, HL60, U937 and K562 cell lines. The effect of in vitro purification was evaluated by the determination of GMCFU and tCFU. RESULTS: Microwave and vincristine showed inhibitory effects on the survival rate of GMCFU and tCFU. The inhibition degree showed the correlation between power and concentration. Within a certain range, microwave and vincristine inhibited the survival rate of tCFU in three cell lines. It was significantly greater than that of GMCFU (P<0.05 or P<0.01). The combined effect of microwave and vincristine on the killing of three cell lines was significantly higher than that of vincristine alone (P<0.05), but not significantly increased on GM. - CFU inhibition (P>0.05). Conclusion The combination of microwave and vincristine for the in vitro purification of leukemia cells can produce synergistic effects, and it is worth further study.
