近交系小鼠遗传质量控制的常用方法有皮肤移植、下颌骨形态分析及生化标志等。这些方法各有其一定特点和不足之处。为了寻找一种比较简便可靠的方法,我们在免疫学方面作了初步尝试,制备了具有品系特异性的多价同种异系抗血清,以检测近交系小鼠的遗传质量。现将其方法和结果报道如下: 原理将不同近交系小鼠的脾脏淋巴细胞混合制成淋巴细胞悬液,注入不同近交系小鼠体内,获得具有品系特异性的多价同种异系抗血清。用这种抗血清同来自被检近交系小鼠体内提取的淋巴细胞,进行微量淋巴细胞毒试验,即可鉴别和检测不同近交系小鼠及其遗传质量是否符合纯系动物的要求。
Common methods for genetic quality control of inbred mice include skin grafts, mandibular morphology analysis and biochemical markers. Each of these methods has its own specific characteristics and shortcomings. In order to find a simple and reliable method, we made a preliminary attempt in immunology to prepare multi-valent allo-antiserum with lineage specificity to detect the genetic quality of inbred mice. The methods and results are reported as follows: Principle The different inbred mice splenic lymphocytes were mixed lymphocyte suspension injected into different inbred mice in vivo, with multi-line allogeneic lineage-specific Antiserum. Using this antisera and the lymphocytes extracted from the inbred mice to be tested for lymphocyte cytotoxicity, different inbred mice can be identified and tested for their genetic quality in accordance with the requirements of purebred animals.
