目的研究川丁特罗体内外抗哮喘活性并对其作用进行组织学验证。方法采用离体豚鼠肺支气管灌流实验考察川丁特罗体外扩张支气管作用;以乙酰胆碱和组胺联合使用作为混合引喘剂,喷雾诱发豚鼠喘息,考察川丁特罗对整体动物的抗急性哮喘发作的作用,并通过对豚鼠肺组织病理学检查,进行组织学验证。结果川丁特罗能够显著抑制组胺致离体豚鼠肺支气管灌流量的减少,IC50为3.99 mg·L-1,作用强度大于阳性药马布特罗(IC50为5.67 mg·L-1)。川丁特罗单剂量(0.125和1 mg·kg-1)灌胃给药,对乙酰胆碱和组胺组成的混合引喘剂喷雾诱发的豚鼠喘息具有一定的保护作用,并能显著改善组织病理学症状。结论川丁特罗体外可明显抑制组胺致豚鼠肺支气管灌流量减少,同时能够有效缓解整体动物急性哮喘症状,显著改善豚鼠肺组织炎症反应。
OBJECTIVE To study the anti-asthmatic activity of chuan-ting luo in vitro and in vivo and its effect on histological verification. Methods Bronchial bronchial perfusion of guinea pig lung was used to investigate the bronchial expansion of chonbuterol in vitro. The combination of acetylcholine and histamine was used as a mixed anti-asthmatic agent to induce guinea pig wheezing by spray. The effect of chrydotrelor on acute asthma attack Through the pathological examination of the lungs of guinea pigs, histological verification. Results Chlamydrotreol significantly inhibited the decrease of bronchoalveolar perfusion in guinea pigs induced by histamine, with an IC50 of 3.99 mg · L-1 and an intensity of 5.67 mg · L-1. Chuan Dingluo single dose (0.125 and 1 mg · kg-1) intragastric administration of acetylcholine and histamine composition of mixed anti-asthmatic agent spray-induced guinea pig wheezing has a protective effect, and can significantly improve histopathology symptom. CONCLUSION Chuan Ding Tro in vitro can inhibit histamine-induced guinea pig lung bronchial perfusion decreased, while can effectively alleviate the overall acute asthma symptoms, significantly improve the guinea pig lung inflammation.
