采用RAPD技术对28株厚壁型和薄壁型巨龙竹样本进行遗传分化研究。结果表明:88条随机引物中筛选出17条重复性好、带型清晰明亮的引物,用于RAPD标记,共检测出131个位点,其中多态性位点118个,多态性程度为90.08%。薄壁型巨龙竹的多态性高于厚壁型巨龙竹,2种类型的遗传分化系数为0.154 6。UPGMA聚类分析显示大部分薄壁型和厚壁型巨龙竹各自聚成一类,证明厚壁型巨龙竹和薄壁型巨龙竹在DNA水平上存在明显差异。
RAPD technique was used to analyze the genetic differentiation of 28 samples of giant-wall and thin-walled dragonfly. The results showed that 17 of 88 random primers were screened for their good repeatability and clear bands. RAPD markers were used to detect 131 loci, of which 118 polymorphic loci were polymorphic 90.08%. The thin-walled giant dragon bamboo polymorphic higher than the thick-walled dragon dragon, two types of genetic differentiation coefficient of 0.154 6. UPGMA cluster analysis showed that most of the thin-walled and thick-walled dragonflies clustered together, which proved that there were significant differences in the DNA levels between the thick-walled dragonfly and the thin-walled dragonfly.
