目的探讨TNF-α对骨巨细胞瘤u-PA系统mRNA表达的调节及意义。方法骨巨细胞瘤原代培养及传代,加入外源性TNF-α,观察加入TNF-α前后细胞生长特性的变化,流式细胞分析仪检测细胞的增殖指数,RTPCR检测u-PA系统mRNA的表达。结果原代培养过程中可见到三种细胞:多核巨细胞(MGC)、巨噬细胞样单核细胞(MC)及纤维母细胞样单核细胞(FC),传代几次以后只有FC及MC;RT-PCR检测细胞中有u-PA、uPAR及PAI-1mRNA表达,加入TNF-α后mRNA的表达较加入前增高(P<0.05);流式细胞分析仪检测细胞的增殖指数(PI),加入TNF-α后PI值较加入前增高(P<0.05)。结论外源性TNF-α促进骨巨细胞瘤原代培养细胞u-PA系统mRNA的表达及细胞增殖。
Objective To investigate the regulatory effect of TNF-α on mRNA expression of u-PA in giant cell tumor of bone. Methods The primary culture and passage of giant cell tumor of bone were treated with exogenous TNF-α, the growth of cells was observed before and after the addition of TNF-α, the proliferation index was detected by flow cytometry, the mRNA of u-PA was detected by RTPCR expression. Results Three kinds of cells were observed in primary culture: multinucleated giant cells (MGC), macrophage - like monocytes (MC) and fibroblast - like mononuclear cells (FC) The expression of u-PA, uPAR and PAI-1 mRNA in the cells was detected by RT-PCR. The mRNA expression of TNF-α and TNF-α were significantly increased (P <0.05). The proliferation index (PI) After adding TNF-α PI increased compared with before (P <0.05). Conclusion Exogenous TNF-α can promote u-PA mRNA expression and cell proliferation in primary cultured human giant cell tumor of bone.
