新采收的山葡萄(VitisamurensisRupr.)种子风干后含水10.92%,硅胶干燥使其含水量分别降至6.2%和3.5%。经过干燥脱水的种子TTC染色百分率同风干种子相比没有差异,均为100%。种子密封置室温、0~5℃、-18℃及-196℃下保存5年。室温下保存的种子全部丧失生活力,低温下保存的种子均有存活,其中以-196℃含水量3.5%的种子出苗率较最高,达74%。试验证明山葡萄种子含水量降至3.5%不损害种子生活力,并可用低温或超低温保存,应确定为正常型种子。
Freshly harvested Vitis amurensis Rupr. Seeds dried 10.92% air dried and dried to a moisture content of 6.2% and 3.5%, respectively. The percentage of TTC stained dried desiccated seeds was 100% compared to air-dried seeds. Seed seal set at room temperature, 0 ~ 5 ℃, -18 ℃ and -196 ℃ for 5 years. The seeds stored at room temperature all lost their viability, and all the seeds preserved at low temperature survived. Among them, the seed germination rate at the water content of -196 ℃ 3.5% was the highest, reaching 74%. Experiments show that the grape seed water content reduced to 3.5% does not affect the viability of seeds, and can be cryogenic or cryogenic preservation, should be identified as normal seed.
