目的:以双黄连为模型药物,对微透析技术应用于中药复方药动学研究进行探索。方法:利用反透析法获得透析液,HPLC梯度洗脱测定透析液中待测成分的含量以计算双黄连体外回收率,考察同一成分在不同成分组成的透析媒介中回收率变化情况,并考察浓度、流速对回收率的影响。结果:随着透析媒介中物质数量的增多,特定成分的回收率有下降趋势,但与透析媒介中该成分的浓度无关,在相同浓度下回收率随流速增大而减少。结论:微透析技术可用于中药复方药动学研究。
Objective: Shuanghuanglian as a model drug, the application of microdialysis technology in traditional Chinese medicine pharmacokinetic study to explore. Methods: The dialysate was obtained by reverse dialysis, and the content of analyte in the dialysate was determined by HPLC gradient elution to calculate the recovery rate of Shuanghuanglian in vitro. The recovery of the same component in the dialysis medium with different components was investigated. The concentration , Flow rate on the recovery rate. Results: With the increase of the amount of substance in the dialysis medium, the recovery rate of the specific components showed a downward trend, but not with the concentration of the components in the dialysis medium. The recovery rate decreased with the increase of the flow rate at the same concentration. Conclusion: Microdialysis technique can be used to study the pharmacokinetics of traditional Chinese medicine.
