目的探讨老年三阴性乳腺癌(TNBC)中PTEN、DJ-1的表达及其临床意义。方法收集本院行手术切除治疗的年龄≥60岁TNBC患者85例,另选择同期年龄≥60岁的正常乳腺组织50例为对照组,采用SP免疫组织化学法检测PTEN、DJ-1蛋白的表达,并分析两者表达的相互作用及其与老年TNBC临床病理特征的关系。结果 PTEN、DJ-1在85例老年TNBC组织中的阳性表达率分别为28.2%(24/85)和74.1%(63/85),在50例正常乳腺组织中的阳性表达率分别为100%(50/50)和6.0%(3/50),老年TNBC组织中DJ-1阳性表达率明显高于正常乳腺组织(P<0.05),而PTEN阳性表达率明显低于正常乳腺组织(P<0.05)。在老年TNBC组织中,TNM分期(Ⅲ~Ⅳ)、伴有腋窝淋巴结转移、伴有脉管癌栓者的PTEN阳性表达率明显低于TNM分期(I~Ⅱ)、无腋窝淋巴结转移、无脉管癌栓者(P<0.05);病理分级(Ⅲ~Ⅳ)、TNM分期(Ⅲ~Ⅳ)、伴有腋窝淋巴结转移、伴有脉管癌栓者的DJ-1阳性表达率明显高于病理分级(I~Ⅱ)、TNM分期(I~Ⅱ)、无腋窝淋巴结转移、无脉管癌栓者(P<0.05)。结论老年TNBC组织中存在PTEN的表达缺失,DJ高表达,且两者均对TNBC患者的淋巴结转移、侵袭性强和容易远处转移的临床病理特征有重要影响。
Objective To investigate the expression of PTEN and DJ-1 in elderly triple negative breast cancer (TNBC) and its clinical significance. Methods Totally 85 TNBC patients aged ≥60 years undergoing surgical resection in our hospital were collected. Another 50 normal breast tissues of the same age≥60 years were selected as the control group. SP immunohistochemistry was used to detect the expression of PTEN and DJ-1 protein , And analyzed the relationship between the expression of the two and its relationship with the clinical and pathological features of elderly TNBC. Results The positive rates of PTEN and DJ-1 in 85 elderly TNBC tissues were 28.2% (24/85) and 74.1% (63/85), respectively. The positive rates of PTEN and DJ-1 in 100 cases of normal breast tissues were 100% (50/50) and 6.0% (3/50). The positive expression of DJ-1 in TNBC tissues was significantly higher than that in normal breast tissues (P <0.05), while the positive expression rate of PTEN was significantly lower than that in normal breast tissues (P < 0.05). In elderly TNBC tissues, the positive expression rates of PTEN in TNM staging (Ⅲ ~ Ⅳ) accompanied with axillary lymph node metastasis and vascular thrombolysis were significantly lower than those in TNM staging (I ~ Ⅱ), without axillary lymph node metastasis, without pulse The positive rate of DJ-1 in pathological grade (Ⅲ ~ Ⅳ), TNM stage (Ⅲ ~ Ⅳ), accompanied with axillary lymph node metastasis and vascular thrombosis was significantly higher than that in the control group (P <0.05) Grading (I ~ Ⅱ), TNM staging (I ~ Ⅱ), no axillary lymph node metastasis, no vascular tumor thrombus (P <0.05). Conclusion There is a lack of expression of PTEN and high expression of DJ in elderly TNBC tissues. Both of them have an important influence on the clinicopathological features of TNBC patients with lymph node metastasis, aggressive invasion and distant metastasis.
