目的检测人髓细胞白血病基因1(MCL-1)蛋白在前列腺癌组织中的表达及其对前列腺癌预后的影响并初步探索其机制。方法利用免疫组织化学方法检测517例前列腺癌组织及癌旁组织中MCL-1蛋白的表达,统计学分析其与前列腺癌患者预后的相关性。干涉前列腺癌细胞系中人MCL-1基因的表达,利用MTT及克隆形成实验检测前列腺癌细胞增殖活性、原位末端转移酶标记技术(TUNEL)检测细胞凋亡能力。结果 MCL-1在前列腺癌组织中的表达显著高于癌旁组织,且MCL-1高表达的前列腺癌患者预后较差。在前列腺癌细胞中干涉MCL-1的表达后,前列腺癌细胞的增殖及抗凋亡能力显著降低。结论MCL-1在前列腺癌组织高表达,干涉前列腺癌细胞中MCL-1表达后可抑制细胞的增殖并促进其凋亡。
Objective To detect the expression of human myeloid leukemia gene 1 (MCL-1) in prostate cancer and its effect on the prognosis of prostate cancer and to explore its mechanism. Methods Immunohistochemistry was used to detect the expression of MCL-1 protein in 517 cases of prostate cancer and its adjacent tissues. The correlation between MCL-1 protein and the prognosis of patients with prostate cancer was statistically analyzed. Interfering with the expression of human MCL-1 gene in prostate cancer cell lines, the proliferation activity of prostate cancer cells was detected by MTT assay and clone formation assay. TUNEL assay was used to detect the apoptotic capacity. Results The expression of MCL-1 in prostate cancer tissues was significantly higher than that in paracancerous tissues, and the prognosis of MCL-1-overexpressing prostate cancer patients was poor. After interference of MCL-1 expression in prostate cancer cells, the proliferation and anti-apoptotic ability of prostate cancer cells are significantly reduced. Conclusion MCL-1 is overexpressed in prostate cancer tissues and the expression of MCL-1 is down-regulated in prostate cancer cells, which may inhibit the proliferation and promote the apoptosis of MCL-1 cells.
