目的:建立一种快速溶剂萃取(ASE)-高效液相色谱检测辣椒中辣椒素和二氢辣椒素的新方法。方法:以甲醇-四氢呋喃(1:1)为萃取剂,考察了萃取温度、萃取时间、萃取循环次数3个因素对ASE萃取效果的影响,随后用高效液相色谱进行检测,并将该方法与国标方法GB/T 21266—2007和药典方法《中国药典》2010版(一部)进行了对照试验。结果:最佳萃取工艺:萃取温度为60℃、萃取时间为20 min、萃取循环次数2次。在此条件下用高效液相色谱法检测了4种辣椒样品中辣椒素和二氢辣椒素的含量,检测结果同国标方法较一致,且优于《中国药典》2010版(一部)。结论:建立的快速溶剂萃取(ASE)-高效液相色谱检测方法具有高效、自动化、无毒、快速的特点,对辣椒中辣椒素和二氢辣椒素的含量检测取得了良好效果,是一种具有实用价值的分析方法。
Objective: To establish a new method of rapid solvent extraction (ASE) - high performance liquid chromatography detection of capsaicin and capsaicin in pepper. Methods: The effects of extraction temperature, extraction time and number of extraction cycles on the extraction efficiency of ASE were investigated by using methanol-tetrahydrofuran (1: 1) as extraction solvent. The effect of ASE extraction was examined by high performance liquid chromatography The national standard method GB / T 21266-2007 and the pharmacopoeia method “Chinese Pharmacopoeia” 2010 edition (a) for the control test. Results: The best extraction process: the extraction temperature was 60 ℃, the extraction time was 20 min, the extraction cycle times 2 times. Under these conditions, the contents of capsaicin and capsaicin in four kinds of pepper samples were detected by HPLC. The results were in good agreement with the national standard method and better than the 2010 edition of Chinese Pharmacopoeia. Conclusion: The established rapid solvent extraction (ASE) - high performance liquid chromatography has the characteristics of high efficiency, automation, non-toxicity and rapidity. The detection of capsaicin and capsaicin in pepper has achieved good results. Practical method of analysis.
