目的:研究葛根通脉饮对高同型半胱氨酸诱导增殖的兔主动脉血管平滑肌细胞内质网应激相关基因免疫球蛋白重链结合蛋白(immunoglobulin heavy chains binding protein,BIP)表达的影响,探讨其抗动脉粥样硬化的机制。方法:30只大耳白兔随机分为空白组、高脂组、可定组(1 mg·kg-1·d-1)、葛根通脉饮低、高剂量组(5,10 g·kg-1·d-1)。空白组给予普通饲料饲喂,高脂组在普通饲料中加入2%胆固醇及0.02%蛋氨酸,可定组在高脂饲料喂养同时加入可定,葛根通脉饮高、低剂量组在高脂饲料的基础上加入不同剂量的葛根通脉饮药物,药物均采用ig给药。饲喂9周后,全自动生化仪测定各组血清总胆固醇(TC),甘油三酯(TG),低密度脂蛋白(LDL),高密度脂蛋白(HDL);胸主动脉切片病理学检查,Western blot法检测BIP蛋白的表达量。结果:与空白组比较,高脂组血清TC,TG,LDL,Hcy水平升高,HDL水平降低(P<0.05,P<0.01);与高脂组相比,可定组及葛根通脉饮两个剂量组血清TC,TG,LDL,Hcy水平降低,HDL水平升高,差异均有统计学意义(P<0.05,P<0.01)。与空白组比较,高脂组主动脉组织BIP蛋白表达明显上调(P<0.01),与高脂组比较,可定及葛根通脉饮两个剂量均能下调主动脉组织BIP蛋白表达(P<0.05)。病理学检验结果显示,可定及葛根通脉饮两个剂量均能明显延缓兔动脉粥样硬化病变进展。结论:葛根通脉饮有抗动脉粥样硬化的作用,其作用机制之一与调控BIP蛋白表达密切相关。
Objective: To study the effect of Gegen Tongmai Drink on the expression of endoplasmic reticulum stress-related gene immunoglobulin heavy chain binding protein (BIP) in rabbit aorta vascular smooth muscle cells (VSMCs) induced by homocysteine, Explore its anti-atherosclerosis mechanism. Methods: Thirty rabbits were randomly divided into blank group, high-fat group, given group (1 mg · kg-1 · d-1) -1 · d-1). The blank group was fed with normal feed, while the high fat group was fed with 2% cholesterol and 0.02% methionine in the normal feed. At the same time, Based on the addition of different doses of Gegen Tongmai drink drugs, drugs are used ig administration. Serum total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL) and high density lipoprotein (HDL) were measured by automatic biochemical analyzer after feeding for 9 weeks. Thoracic aortic biopsy Western blot was used to detect the expression of BIP protein. Results Compared with the blank group, the levels of TC, TG, LDL and Hcy in high fat diet group increased and the HDL level decreased (P <0.05, P <0.01). Compared with the high fat diet group, Serum levels of TC, TG, LDL and Hcy decreased and HDL levels increased in two dose groups (P <0.05, P <0.01). Compared with the blank group, the BIP protein expression in the aorta of the high-fat diet group was significantly increased (P <0.01). Compared with the high-fat diet group, the Puerarin Tongmai drink could both reduce the BIP protein expression in the aorta tissue (P < 0.05). The results of pathology test showed that both doses of Gegen Tongmai drink could delay the development of atherosclerotic lesions in rabbits. Conclusion: Gegen Tongmai has the anti-atherosclerotic effect, and one of its mechanisms is closely related to the regulation of BIP protein expression.
