[目的]通过多位点连锁分析寻找妊娠糖尿病(GDM)的易感基因。[方法]采用非参数连锁统计分析法,以101对妊娠糖尿病患病同胞对DNA为研究对象,在支持连锁位点D10S587附近区域加密微卫星标记(D10S217、D10S583、D10S1213、D10S1230、D10S1237)做多位点连锁分析。[结果]结果显示:与D10S587同样位于10q26且位置非常接近的位点D10S1213表现为与GDM易感基因呈显著连锁(NPL-Score=2.122,P=0.0169)。[结论]具有2型糖尿病致病基因的10q26区域同样具有致GDM的基因。它们很可能是同一基因。
[Objective] To find the susceptible genes of gestational diabetes mellitus (GDM) by multi-locus linkage analysis. [Method] Non-parametric linkage statistical analysis was used to study the DNA of 101 siblings with gestational diabetes mellitus, and more microsatellite markers (D10S217, D10S583, D10S1213, D10S1230 and D10S1237) were constructed in the vicinity of D10S587 supporting the linkage sites Site linkage analysis. [Result] The results showed that D10S1213 showed a significant linkage to the GDM susceptibility gene (NPL-Score = 2.122, P = 0.0169) at the position 10q26 and closely located to D10S587. [Conclusion] The region of 10q26 with pathogenicity of type 2 diabetes also has the gene of GDM. They are likely to be the same gene.
