目的研究子宫内膜癌组织中乏氧诱导因子-1α(HIF-1α)与血管内皮生长因子(VEGF)的表达情况及其与肿瘤血管生成的关系。方法选取2012年2月至2015年12月河北工程大学附属医院收治的48例子宫内膜癌患者(内膜癌组),应用免疫组织化学法分别检测48例子宫内膜癌组织及15例正常子宫内膜(正常内膜组)组织中HIF-1α和EGF的表达情况,并用CD34抗体标记血管内皮细胞,测定微血管密度(MVD)值。对子宫内膜癌组织中HIF-1α、VEGF表达与临床参数及MVD进行相关性分析。结果内膜癌组患者组织中HIF-1α和VEGF高表达率分别为54.17%、75.00%,均明显高于正常内膜组的6.67%、13.33%,组间比较差异均有显著统计学意义(P<0.01);内膜癌组与正常内膜组组织的MVD计数分别为(35.51±21.36)条、(3.66±4.52)条,组间比较差异有显著统计学意义(P<0.01);内膜癌组织中HIF-1α和VEGF表达与患者的年龄、临床分期、肌层浸润程度无关(P>0.05),与组织分化程度相关(P<0.05),而MVD与患者年龄、组织分化程度、临床分期、肌层浸润程度均有相关性(P<0.05)。HIF-1α与VEGF的表达呈正相关(r=0.485,P=0.021);HIF-1α与MVD呈正相关(r=0.680,P=0.045);VEGF和MVD亦呈正相关(r=0.423,P=0.035)。结论子宫内膜癌组织中HIF-1α和VEGF均有高水平表达,并与肿瘤血管生成密切相关,两者可作为子宫内膜癌的肿瘤标志物,为临床抗肿瘤治疗提供新的方向。
Objective To investigate the expression of hypoxia inducible factor-1α (HIF-1α) and vascular endothelial growth factor (VEGF) in endometrial carcinoma and its relationship with tumor angiogenesis. Methods Forty-eight patients with endometrial carcinoma (endometrial carcinoma) who were treated in Affiliated Hospital of Hebei Engineering University from February 2012 to December 2015 were enrolled in this study. Forty-eight cases of endometrial carcinoma and 15 cases of normal endometrial carcinoma were detected by immunohistochemistry The expression of HIF-1α and EGF in endometrium (normal endometrium) tissues was detected. Vascular endothelial cells were labeled with CD34 antibody and the microvessel density (MVD) was measured. The correlation between the expression of HIF-1α, VEGF, clinical parameters and MVD in endometrial carcinoma was analyzed. Results The high expression rates of HIF-1α and VEGF in endometrial carcinoma patients were 54.17% and 75.00%, respectively, which were significantly higher than 6.67% and 13.33% of those in normal endometrium, respectively. There were significant differences between the two groups (P <0.01). The MVD counts in endometrial carcinoma group and normal endometrium group were (35.51 ± 21.36) and (3.66 ± 4.52), respectively. There was significant difference between the two groups (P <0.01) The expression of HIF-1α and VEGF was not related to the age, clinical stage and myometrial infiltration (P> 0.05), but correlated with the degree of differentiation (P <0.05). However, the correlation between MVD and age, histological differentiation, Clinical stage and myometrial invasion were correlated (P <0.05). There was a positive correlation between the expression of HIF-1α and VEGF (r = 0.485, P = 0.021). There was a positive correlation between HIF-1α and MVD (r = 0.680, ). Conclusion The expression of HIF-1α and VEGF in endometrial carcinoma are highly expressed and closely related to tumor angiogenesis, both of which can be used as tumor markers of endometrial cancer, providing a new direction for clinical anti-tumor therapy.
