目的探讨在乳头状甲状腺癌K1及MCF7细胞系中,雌激素(E2)对hTERT的mRNA及蛋白表达的影响。方法应用Western Blot检测乳头状甲状腺癌K1细胞中雌激素受体(ER)的表达;应用共聚焦激光扫描显微镜观察ER在E2刺激后定位的改变;应用Real-time PCR检测E2刺激对人端粒酶逆转录酶(hTERT)mRNA表达的影响;应用Western Blot检测E2刺激对hTERT的蛋白表达的影响。结果 K1细胞系中存在ERα阳性表达,在E2刺激后阳性表达产物入核,E2刺激能够促进K1细胞中hT ERTmRNA和蛋白表达水平上调。结论雌激素促进hT ERT在乳头状甲状腺癌细胞中的表达。
Objective To investigate the effect of estrogen (E2) on mRNA and protein expression of hTERT in papillary thyroid carcinoma K1 and MCF7 cell lines. Methods The expression of estrogen receptor (ER) in papillary thyroid carcinoma K1 cells was detected by Western Blot. The localization of estrogen receptor (ER) was observed by confocal laser scanning microscopy. The effect of E2 stimulation on the expression of human telomeres Enzyme reverse transcriptase (hTERT) mRNA expression; application of Western Blot detection of E2 stimulation hTERT protein expression. Results The positive expression of ERα in K1 cell line was observed. The positive expression of ERα in the K1 cell line was found in the nucleus. E2 stimulation promoted the up-regulation of hT ERT mRNA and protein in K1 cell line. Conclusion Estrogen enhances the expression of hT ERT in papillary thyroid carcinoma cells.
