目的评价丘脑底核混沌电刺激对大鼠癫疒间发作的影响并探讨其作用的可能机制。方法SD大鼠随机分3组:电刺激组、假刺激组和对照组。(1)电刺激组丘脑底核植入电极给予混沌电信号预刺激1周,美解眠腹腔注射诱发强直-阵挛发作,记录发作不同阶段发作行为、脑电图间疒性放电的潜伏期和点燃后发作总的持续时间;(2)假刺激组除不接受电刺激外,处理同刺激组;(3)对照组不植入电极,不接受电刺激。采用免疫组织化学SABC法观察大鼠顶叶皮质和海马CA1、CA3区γ-氨基丁酸(GABA)免疫反应细胞的改变。结果丘脑底核混沌电刺激明显延长大鼠癫间疒Ⅰ、Ⅴ级发作的潜伏期,缩短点燃后发作的总持续时间,与脑电图获得结果一致。电刺激组皮质和海马CA1、CA3区GABA免疫反应阳性细胞多于对照组。假刺激组结果类似,作用弱于电刺激组。结论丘脑底核混沌电刺激对大鼠癫间疒发作有一定保护效应,其机制可能与皮层及海马增强GABA的抑制有关。
Objective To evaluate the effect of subthalamic nucleus electrical stimulation on epileptic seizures in rats and to explore its possible mechanism. Methods SD rats were randomly divided into 3 groups: electrical stimulation group, sham stimulation group and control group. (1) In the stimulation group, the electrodes of the subthalamic nucleus were pre-stimulated for 1 week by chaotic electrical signals. The mice in the treatment group were given intraperitoneal injection of tetanus to induce tonic-clonic seizures. The seizure behavior, (2) In the sham stimulation group, the same stimulation group was not treated except for the electrical stimulation; (3) The control group did not implant the electrodes and did not receive the electrical stimulation. Immunohistochemical SABC method was used to observe the changes of γ-aminobutyric acid (GABA) immunoreactive cells in parietal cortex and hippocampal CA1 and CA3 areas. Results Chaotiarigal nucleus electrical stimulation significantly prolonged the episodes of epilepsy Ⅰ and Ⅴ episodes in rats and shortened the total duration of episodes after ignition, consistent with the results obtained by electroencephalography. The number of GABA immunoreactive cells in cortex and hippocampus CA1 and CA3 area in electrical stimulation group was more than that in control group. The sham stimulation group had similar results and weaker effects than the electrical stimulation group. Conclusions The chaotroic electrical stimulation of the subthalamic nucleus has a protective effect on epileptic seizures in rats. The mechanism may be related to the inhibition of GABA in the cortex and hippocampus.
