目的 :研究抗 RAP抗体对肾小球上皮足突细胞 RAP基因表达调控。方法 :受体相关蛋白 (RAP)多抗 ,作用于体外分离培养的 SD大鼠肾小球上皮足突细胞 12小时 ,以模拟 Heym ann肾炎 (HN)模型体内循环抗体作用于肾小球发病过程 ,采用 RT- PCR半定量分析的方法研究肾小球足突细胞受体相关蛋白基因表达。结果 :RAP多抗作用于培养肾小球上皮足突细胞使得细胞 RAP基因表达增高。结论 :RAP循环抗体调控肾小球上皮足突细胞 RAP基因表达增加并导致 RAP表达增加及抗原抗体复合物不断堆积是 Heymann肾炎的分子机制之一。
Objective: To study the regulation of RAP gene expression in glomerular epithelial cells by anti-RAP antibodies. METHODS: Polyclonal antibody against receptor-associated protein (RAP) was used to effect the in vitro differentiation of cultured glomerular glial cells from cultured SD rats for 12 hours in order to mimic the circulating antibody in the HeNa ann nephritis (HN) model in the pathogenesis of glomeruli , RT-PCR semi-quantitative analysis of glomerular podocyte receptor-related protein gene expression. Results: RAP multi-antibody treatment of glomerular epithelial cells in the footprotein cells RAP gene expression increased. Conclusion: RAP circulating antibody regulates the expression of RAP gene in glomerular epithelial cells and leads to the increase of RAP expression and the accumulation of antigen-antibody complex. It is one of the molecular mechanisms of Heymann’s nephritis.
