目的建立高效毛细管电泳法分离苯丙哌林、益康唑、酚苄明和美西律共4种手性药物对映体。方法采用未涂层的熔融毛细管柱,以磺酸化-β-环糊精(sulfated-β-cyclodextrin,S-β-CD)为手性选择剂,背景电解质选用磷酸盐缓冲液,分离电压为20 k V。分别考察了手性选择剂S-β-CD的质量浓度、背景电解质p H值、缓冲溶液浓度和温度对对映体分离的影响,优化了分离条件。结果在最佳条件下,4种手性药物对映体均得到完全分离,最佳分离度(R)分别为6.4、4.2、14.7和3.1。结论 S-β-CD对4种碱性药物均具有较高的对映体选择性。
OBJECTIVE To establish a high performance capillary electrophoresis method for the separation of enantiomers of 4 chiral drugs, benproperine, econazole, phenoxybenzamine and mexiletine. Methods Uncoated fused silica capillary column was used to select sulfated-β-cyclodextrin (S-β-CD) as chiral selector and phosphate buffer as background electrolyte. The separation voltage was 20 k V. The effects of chiral selector S-β-CD concentration, background electrolyte p H value, buffer concentration and temperature on the separation of enantiomers were investigated respectively, and the separation conditions were optimized. Results Under the optimal conditions, the enantiomers of the four chiral drugs were completely separated. The optimal resolution (R) was 6.4, 4.2, 14.7 and 3.1 respectively. Conclusion S-β-CD has high enantioselectivity for all four basic drugs.
