目的 探讨小鼠骨髓树突状细胞(dendritic cell,DCs),体外经Hepa1-6肝癌细胞株抗原粗提物致敏后,对特异性细胞毒 T淋巳细胞(CTL)的诱导作用。方法 自小鼠骨髓分离 DCs前体细胞,经 GM-CSF+IL-4培养、扩增;制备 Hepa 1-6小鼠肝癌细胞株抗原粗提物,体外脉冲DCs,检测DCs诱导同基因型小鼠T细胞增殖能力及特异性CTL的反应能力。结累和结论 经肿瘤抗原致敏的DCs组织相容性分子(MHC-Ⅰ、Ⅱ)及共刺激分子(B7-2、B7-1)表达明显增高,能诱导较强的同基因型小鼠T细胞增殖.且能诱导Hepa 1-6特异性细胞毒T淋巴细胞。
Objective To investigate the induction of specific cytotoxic T lymphocytes (CTLs) by sensitization of mouse bone marrow dendritic cells (DCs) after in vitro sensitization with Hepa 1-6 liver cancer cell strains. METHODS: DCs precursor cells were isolated from mouse bone marrow and cultured and amplified by GM-CSF + IL-4. Hepa 1-6 mouse liver cancer cell strains were prepared with crude extracts, DCs were pulsed in vitro, and DCs were induced to induce syngeneic mice. Cell proliferation and specific CTL response. Conclusions and Conclusions The expression of histocompatibility molecules (MHC-I, II) and co-stimulatory molecules (B7-2, B7-1) in DCs sensitized with tumor antigens is significantly increased, and can induce strong isogenic mice. T cell proliferation. And can induce Hepa 1-6 specific cytotoxic T lymphocytes.
