用地高辛(digoxigenin-11-dUTP)标记杜氏利什曼原虫kDNA重组片段作为探针,与不同种株利什曼原虫kDNA斑点杂交,获得杜氏利什曼原虫一种特异性kDNA片段,双脱氧核苷酸链终止法测定该片段长321bp,计算机检索及同源性分析比较再次表明该片段具有较高的种特异性。该片段可作为探针用于利什曼病病原体的检测,有助于虫株鉴定,也可以作为聚合酶链反应(PCR)扩增的靶序列。
Using digoxigenin-11-dUTP-labeled kDNA recombination fragment of Leishmania donovani as a probe to hybridize with different species of Leishmania kDNA spots to obtain a specific kDNA fragment of Leishmania donovani, dideoxy The fragment length was 321bp determined by nucleotide chain termination method. Comparison of computer search and homology analysis again showed that the fragment was highly species-specific. The fragment can be used as a probe for the detection of leishmaniasis pathogens, identification of strains, and as a target for polymerase chain reaction (PCR) amplification.
