目的:观察同型半胱氨酸(Homocysteine,Hcy)对大鼠脑微血管内皮细胞(Ratcerebralmicrovascularendothelialcecl,RCMEC)的损伤效应,以揭示Hcy致动脉粥样硬化的可能机制。方法:1将不同浓度的Hcy分别加入培养的RCMEC中孵育2、4、6、8、10h后,测定细胞乳酸脱氢酶(LDH)释放率;2观察同型半胱氨酸损伤组和对照组细胞生长曲线以及相差显微镜下、透射电镜下结构改变。结果:Hcy使RCMEC的LDH释放率明显增加,破坏细胞形态结构并抑制其生长。结论:Hcy可能通过氧化应激损伤机制对RCMEC产生毒性损伤作用。
Objective: To observe the effect of Homocysteine (Hcy) on rat cerebral microvascular endothelial cell (RCMEC) in order to reveal the possible mechanism of Hcy-induced atherosclerosis. Methods: 1 Hcy of different concentrations were added to cultured RCMEC respectively and incubated for 2, 4, 6, 8 and 10 h, then the cell lactate dehydrogenase (LDH) release rate was measured. 2 Homocysteine-induced injury group and control group Cell growth curve and phase contrast microscope, TEM changes in structure. Results: Hcy significantly increased the release rate of LDH from RCMEC, disrupted the morphological structure of cells and inhibited their growth. Conclusion: Hcy may induce toxic injury to RCMEC through oxidative stress injury mechanism.
