以原代培养乳大鼠心肌细胞的搏动和乳酸脱氢酶活性为指标,评估无钙和重补钙培养后其细胞的损伤变比。无钙培养后,心肌细胞搏动变得不规则、少数细胞停搏,酶活性降低无显著差异(P>0.05);无钙后重补钙,细胞完全停搏,酶活性显著降低(P<0.05).结果进一步证实了培养心肌细胞的钙矛盾损伤效应。
The pulsatility and lactate dehydrogenase activity of primary cultured rat myocardial cells were used as indexes to evaluate the injury ratio of cells cultured in calcium-free and calcium-supplemented medium. After calcium-free culture, the beating of cardiomyocytes became irregular and the number of cardioplegia and enzyme activity decreased significantly (P> 0.05). After calcium-free calcium supplementation, cells were completely arrested and the activity of enzyme was significantly decreased (P <0.05 ) The results further confirmed the calcium contractile damage effect of cultured cardiomyocytes.
