用体外和体内试验,在电镜下比较观察SD大鼠脾树状突细胞(DC)和巨噬细胞(Mφ)的吞噬功能。体外试验以白色念珠菌(CA)、公鸡红细胞(CRBC)和经大鼠抗CRBC抗血清调理的CRBC(OCRBC)为吞噬物,分别加入新分离的含DC和Mφ的脾细胞悬液中,37℃培养1h。DC一般无吞噬现象,而大多数Mφ摄入数量不等的CA、CRBC或OCRBC。体内试验将印度墨汁(碳粒、CC)和热灭活的CA(HKCA),分别经静脉注入动物体内,18h后取脾制成悬液,DC一般也无吞噬现象,仅有些细胞摄入少量细小CC;而大多数Mφ摄入数量不等的CC或HKCA。表明在同一条件下,对非免疫原性和免疫原性物质,脾Mφ均显示了活跃的吞噬功能,在有抗体包被的情况下更为显著;脾DC则无吞噬或仅有微弱的吞噬功能。DC和Mφ吞噬功能的显著不同也证明它们是两种不同的细胞。
The phagocytosis of splenic dendritic cells (DCs) and macrophages (Mφ) in SD rats was observed by electron microscopy in vitro and in vivo. In Vitro Assays Freshly isolated suspension of DCs and Mφ-containing splenocytes was added as a phagocytosis with C. albicans (CA), rooster red blood cells (CRBC) and CRBC (OCRBC) conditioned with rat anti-CRBC antiserum respectively ℃ for 1 h. DCs generally have no phagocytosis, whereas most Mφs ingest CA, CRBC or OCRBC in varying amounts. In vivo, Indian ink (carbon particles, CC) and heat-inactivated CA (HKCA) were injected intravenously into animals respectively. After 18 hours, the spleen was made into a suspension. DC generally did not phagocytose. Small CC; whereas most Mφ intake of CC or HKCA ranging in number. The results showed that under the same conditions, the non-immunogenic and immunogenic substances, spleen Mφ showed active phagocytosis, with antibody coating is more significant; spleen DC is no phagocytosis or only weak phagocytosis Features. Significant differences in the phagocytosis of DC and Mφ also prove that they are two different cells.
