目的 探讨不同生长特性的人宫颈癌细胞亚克隆株CS0 3和CS0 7与其细胞周期相关基因的关系。方法 采用流式细胞仪对两株细胞进行细胞周期分析 ,用含有 2 34条人类全长细胞周期相关基因的cDNA表达芯片 ,对两个同一来源、生长特性不同的人宫颈癌细胞亚克隆株CS0 3、CS0 7进行细胞周期相关基因的差异表达谱分析。结果 正常生长条件下培养的CS0 3细胞G1期百分比明显较CS0 7细胞高 ,而CS0 7细胞S期百分比则较CS0 3细胞有所增加 ;在无血清培养条件下CS0 3细胞在4 8h后凋亡 (亚G1期 )细胞百分比明显高于CS0 7细胞 ,并且随着时间的延长而增加。两个亚克隆细胞株CS0 3、CS0 7有 3个差异表达基因 ,分别为人BN5 1mRNA、人热休克蛋白 90 (hsp90 )mRNA和Mcl 1基因 ,荧光强度的比值分别是 0 4 80、0 4 79和 0 4 90。结论 不同生长特性的宫颈癌细胞亚克隆株与其细胞周期相关基因BN5 1、hsp90和Mcl 1有密切关系
Objective To investigate the relationship between the expression of CS0 3 and CS0 7 and their cell cycle related genes in human cervical cancer cells with different growth characteristics. Methods Flow cytometry was used to analyze the cell cycle of two cell lines. Two cDNA sub-clones of human cervical cancer cell line CS0 with different origin and growth characteristics were obtained by cDNA microarray with 2 34 full-length human cell cycle related genes 3, CS0 7 differential expression profiling of cell cycle related genes. Results The percentage of G1 phase in CS0 3 cells cultured in normal condition was significantly higher than that in CS0 7 cells and the percentage of S phase in CS0 7 cells was increased compared with that in CS0 3 cells. CS0 3 cells in 48 h after culture in serum-free medium The percentage of cells in the sub-G1 phase was significantly higher than that in the CS0 7 cells, and increased with time. The two subclone cell lines CS0 3 and CS0 7 had three differentially expressed genes, namely human BN5 1 mRNA, human HSP90 (Msp 1) gene and Mcl 1 gene respectively. The ratios of fluorescence intensities were 0 4 80 and 0 4 79 And 0 4 90. Conclusion The subclone of cervical cancer cells with different growth characteristics is closely related to the cell cycle related genes BN5 1, hsp90 and Mcl 1
