目的在于探讨IHC-72(3,6-2甲氨基一二苯并碘杂六环依地酸盐)对血管内皮细胞的保护作用。方法将大耳白兔21只随机分为对照(n=5)、实验(n=9)及IHC-72(n=7)3组。利用饲菌复合失血性休克及再灌注损伤的方法和标准复制家兔MODS模型,达标者称为MODS组。IHC-72组于实验前0.5h肌注 IHC-72(0.25ml/kg);对照组不饲菌、不放血、不注药。实验过程中观察血管内皮细胞形态改变及相关的功能指标。结果 MODS对血管内皮细胞明显损伤,质膜下空泡增多,核染色质边集,线粒体和内质网等减少,严重者坏死、脱落。而IHC-72组则内皮细胞损伤轻微,其72h血浆ⅧR∶Ag浓度(114.7±5.9%)与实验前(91.4±0.8%)无显著差异(P<0.05),但与MODS组(167,0±6,1%)有明显差异(P<);ihC-72组72h血浆 6-K-PGFα浓度( 6210± 18. 4pg/ml)与实验前比(643. 7±20. 7)无显著差异(P>0.05),而与 MODS组(467, 1± 14. 2)比有显著差异(P<0.05)。结论以上 IHC-72对MODS血管内皮细胞有一定保护作用。本研究还说明钙
The purpose of this study was to investigate the protective effects of IHC-72 (3,6-2-methyldibenzopyrimidine iodide) on vascular endothelial cells. Methods Twenty-one rabbits were randomly divided into control group (n = 5), experimental group (n = 9) and IHC-72 group (n = 7) MODS model was used to replicate the MODS model of rabbits by using the methods and standards of hemorrhagic shock and reperfusion injury in rabbits. The IHC-72 group was intramuscularly injected with IHC-72 (0.25ml / kg) 0.5h before the experiment; the control group did not feed the bacteria, no bloodletting and no injection. During the experiment, we observed the morphological changes of vascular endothelial cells and related functional indexes. Results MODS significantly damaged vascular endothelial cells, increased vacuolation under the plasma membrane, decreased nuclear chromatin margins, mitochondria and endoplasmic reticulum, and severe necrosis and shedding. The IHC-72 group showed slight damage to endothelial cells, and there was no significant difference in the concentration of plasma ⅧR: Ag between the two groups (114.7 ± 5.9% vs 91.4 ± 0.8%, P <0.05 (P <0.01), but it was significantly different from that of MODS group (167.0 ± 6.1%) (P <0.01).
