目的研究四君子汤总多糖(SJZPS)对化疗荷瘤小鼠脾脏、派氏结(PPs)、肠系膜淋巴结(MLNs)淋巴细胞的影响。方法将BALB/c小鼠随机分为对照组、肿瘤模型组,环磷酰胺(CY)化疗组和四君子汤总多糖治疗(CY+SJZPS)组。CY组和CY+SJZPS组每天腹腔注射CY20 mg.kg-1,CY+SJZPS组同时灌胃SJZPS 1g.kg-1,连续给药9 d。应用流式细胞术检测脾脏、小肠PPs和MLNs中T、B淋巴细胞比例、活化水平以及T细胞亚群的变化。结果 SJZPS能提高肠道PPs中B淋巴细胞的比例和活化水平,降低MLNs中CD4+/CD8+的比值,改善化疗所致的肠道黏膜免疫损伤;SJZPS能提高化疗荷瘤小鼠脾脏淋巴细胞中B淋巴细胞的比例,及T、B淋巴细胞的活化水平,同时降低CD4+/CD8+的比值以改善化疗荷瘤小鼠系统免疫功能,其中SJZPS对B细胞的作用更加明显。结论 SJZPS能有效拮抗化疗药物所致的小鼠肠黏膜免疫功能损伤,同时对整体免疫功能具有改善作用。
Objective To study the effects of SJZPS on spleen lymphocytes, spleen lymph nodes (PPs) and mesenteric lymph nodes (MLNs) in chemotherapy-bearing mice. Methods BALB / c mice were randomly divided into control group, tumor model group, cyclophosphamide (CY) chemotherapy group and Sijunzi Decoction total polysaccharide treatment group (CY + SJZPS). The CY group and the CY + SJZPS group were intraperitoneally injected with CY20 mg.kg-1, while the CY + SJZPS group was administered with 1 g.kg-1 of SJZPS simultaneously for 9 days. Flow cytometry was used to detect the ratio of T and B lymphocytes in spleen, small intestine PPs and MLNs, the activation level and T cell subsets. Results SJZPS could increase the ratio and activation of B lymphocytes in intestinal PPs, decrease the ratio of CD4 + / CD8 + in MLNs, and improve the intestinal mucosal immunologic injury induced by chemotherapy. SJZPS can increase the ratio of B Lymphocyte ratio, and T, B lymphocyte activation level, while reducing the ratio of CD4 + / CD8 in order to improve the immune system of mice with chemotherapy-induced immune function, which SJZPS B cells more obvious. Conclusion SJZPS can effectively prevent the damage of intestinal mucosal immune function induced by chemotherapeutic drugs and improve the overall immune function.
