目的研究光枝勾儿茶全株中的化学成分,尤其是anthraquinone-benzisochromanquinone二聚体成分,探讨其对乙酰胆碱酯酶的抑制活性。方法 采用硅胶柱色谱,Sephadex LH-20凝胶柱色谱进行分离纯化,通过波谱学方法鉴定化合物结构,并对化合物1~3进行体外抗乙酰胆碱酯酶活性粗筛实验。结果从光枝勾儿茶乙醇提取物中分离得到4个anthraquinone-benzisochro-manquinone二聚体化合物,分别鉴定为floribundiquinones A(1),floribundiquinones B(2),floribundiquinones C(3),floribundiquinoneD(4);7个其他已知化合物:阿魏酸(5)、大黄素甲醚-8-O-β-D-葡萄糖苷(6)、大黄素-8-O-β-D-葡萄糖苷(7)、齐墩果酸(8)、钝叶素(9)、木犀草素-4′-O-β-D-葡萄糖苷(10)、单棕榈酸甘油酯(11)。1~3化合物粗筛结果表明,当样品质量浓度为0.6 mg.mL-1时,化合物1和3没有抗乙酰胆碱酯酶活性,而化合物2对乙酰胆碱酯酶的抑制率为57.8%。结论化合物1~4为首次从该植物中分离得到,化合物5~11均为首次从该属植物中发现;其中化合物2对乙酰胆碱酯酶有一定的抑制活性。
OBJECTIVE To study the chemical constituents in the whole plant, especially the anthraquinone-benzisochromanquinone dimer, and to study its inhibitory activity on acetylcholinesterase. Methods The compounds were isolated and purified by column chromatography on silica gel and Sephadex LH-20 gel column chromatography. The structures of compounds were identified by spectroscopic methods. Compounds 1 ~ 3 were screened by anti-acetylcholinesterase activity in vitro. Results Four anthraquinone-benzisochro-manquinone dimer compounds were isolated from the ethanol extract of Cortex Khotanica and identified as floribundiquinones A (1), floribundiquinones B (2), floribundiquinones C (3) and floribundiquinone D (4), respectively. Seven other known compounds were ferulic acid (5), physcion-8-O-β-D-glucoside (6), emodin-8-O-β-D-glucoside Oleanolic acid (8), bluntroptin (9), luteolin-4’-O-β-D-glucoside (10) and glycerol monopalmitate (11). The results of coarse screening of 1 ~ 3 compounds showed that compounds 1 and 3 had no anti-acetylcholinesterase activity when the sample mass concentration was 0.6 mg.mL-1, while the inhibitory rate of compound 2 on acetylcholinesterase was 57.8%. Conclusion Compounds 1 ~ 4 were isolated from this plant for the first time. Compounds 5 ~ 11 were found from this genus for the first time. Among them, compound 2 showed some inhibitory activity against acetylcholinesterase.
