目的:检测非编码RNA FENDRR长链在宣威肺腺癌XWLC-05株细胞与非小细胞肺癌(NSCLC)A549株细胞中的表达,及其对细胞增殖、迁移及侵袭的影响。方法:实时荧光定量PCR检测FENDRR基因在XWLC-05与A549细胞中的表达;构建过表达p EX-3-FENDRR质粒并转染XWLC-05细胞,慢病毒干扰LV3-FENDRR载体转染A549细胞,实时荧光定量PCR检测转染后XWLC-05和A549细胞内FENDRR的表达;MTS法检测细胞增殖;Transwell小室法检测细胞迁移及侵袭能力的变化。结果:XWLC-05细胞中FENDRR基因表达明显较A549细胞低,成功提高转染p EX-3-FENDRR的XWLC-05细胞和降低转染LV3-FENDRR质粒的A549细胞中FENDRR m RNA水平。与对照组相比,过表达FENDRR可明显抑制XWLC-05细胞增殖[(1.23±0.13)vs(1.46±0.25),P<0.05]并降低其迁移、侵袭能力(P<0.05);干扰FENDRR表达,能够促进A549细胞增殖[(0.85±0.02)vs(0.77±0.08),P<0.05]、迁移与侵袭能力(P<0.05)。结论:lnc RNA FENDRR表达下调与宣威肺腺癌的发生发展相关,其在肿瘤恶性化过程中抑制肿瘤细胞的增殖与转移。
OBJECTIVE: To detect the expression of non-coding FENDRR long chain in XWLC-05 Strain and non-small cell lung cancer (NSCLC) A549 cells and its effect on cell proliferation, migration and invasion. METHODS: The expression of FENDRR gene in XWLC-05 and A549 cells was detected by real-time fluorescence quantitative PCR. The plasmid pEX-3-FENDRR was overexpressed and transfected into XWLC-05 cells. A549 cells were transfected with lentivirus vector LV3-FENDRR. The expression of FENDRR in XWLC-05 and A549 cells was detected by real-time fluorescence quantitative PCR. The cell proliferation was detected by MTS assay. The changes of cell migration and invasion were detected by Transwell chamber assay. Results: The FENDRR gene expression in XWLC-05 cells was significantly lower than that in A549 cells. The expression of FENDRR m RNA in XWLC-05 cells transfected with pEX-3-FENDRR and the A549 cells transfected with LV3-FENDRR plasmids were successfully increased. Compared with the control group, the overexpression of FENDRR significantly inhibited the proliferation of XWLC-05 cells (1.23 ± 0.13 vs 1.46 ± 0.25, P <0.05) and decreased the migration and invasion ability (P <0.05) , Which could promote the proliferation of A549 cells [(0.85 ± 0.02) vs (0.77 ± 0.08), P <0.05], migration and invasion ability (P <0.05). Conclusion: The downregulation of lnc RNA FENDRR is associated with the occurrence and development of lung adenocarcinoma of Xuanwei, which inhibits the proliferation and metastasis of tumor cells in the process of malignant tumor.
