目的 观察酸灌注对兔食管下括约肌 (L ES)形态及功能的影响 .方法 采用 0 .1mol·L- 1 HCL (1m L· min- 130 min· d- 1 for 4d )连续行兔食管灌注 ,测定 L ES的压力变化 .以 Fluo- 3作为钙指示剂 ,用激光共聚焦显微镜测定酸灌注前后细胞内游离钙的浓度变化 ,以及其对乙酰胆碱作用(Ach)的不同反应 .结果 酸灌注后 L ES产生明显的炎症改变 ,L ES压力降低 ,L ES平滑肌细胞内游离钙的浓度 [Ca2 + ]i明显降低 (P<0 .0 1) ,在应用 ACh时正常对照组 L ES平滑肌 [Ca2 + ]i 明显升高 (P<0 .0 1) ,而酸灌注组则无明显变化(P>0 .0 5 ) .结论 酸灌注可使兔 L ES产生明显的炎症改变 ,其平滑肌 [Ca2 + ]i 明显降低 ,可能影响兔 L ES平滑肌细胞内储存钙的释放
Objective To observe the effect of acid perfusion on the morphology and function of lower esophageal sphincter (LES) in rabbits.Methods The rabbits were consecutively administered with 0.1 mol·L-1 HCl (1 m L · min-130 min · d-1 for 4d) The change of LES pressure was measured.The change of intracellular free calcium concentration before and after acid perfusion and the different reaction to acetylcholine (Ach) were measured by laser confocal microscopy with Fluo-3 as the calcium indicator.Results After acid perfusion L ES induced obvious inflammatory changes, L ES pressure decreased, L ES smooth muscle cells intracellular free calcium concentration [Ca2] i significantly reduced (P <0.01), ACh when normal control group L ES smooth muscle [Ca2 + ] i significantly increased (P <0.01), while there was no significant difference in acid perfusion group (P> 0.05) .Conclusion Acid perfusion can induce a significant inflammatory change in L ES in rabbits, and its smooth muscle [Ca2 + ] i significantly reduced, may affect the release of stored calcium in rabbit L ES smooth muscle cells
