应用顺铂诱导体外培养的人食管癌细胞发生凋亡,在光镜下观察细胞的形态学改变,用免疫细胞化学方法染色以检测肿瘤细胞白介素1β转化酶(ICE)基因的表达情况。结果显示,顺铂处理后的肿瘤表现为出芽、与相邻细胞分离、核碎裂、凋亡小体形成及其被吞噬现象。ICE蛋白在未用药处理的对照细胞呈阴性表达,而发生凋亡的细胞及凋亡小体呈阳性表达,呈阳性表达的细胞数目及着色强度随药物作用时间延长而增加,然而一些不具备凋亡形态特征或处于凋亡前状态的肿瘤细胞ICE蛋白也呈阳性表达。结果表明ICE基因可能参与了顺铂诱导肿瘤细胞凋亡的调节,ICE蛋白在顺铂诱导肿瘤细胞凋亡的早期阶段可能起着一定作用
Apoptosis of human esophageal cancer cells was induced by cisplatin in vitro. The morphological changes of the cells were observed under light microscope. The expression of interleukin 1β-converting enzyme (ICE) gene in tumor cells was detected by immunocytochemistry. The results showed that cisplatin-treated tumors exhibited budding, separation from adjacent cells, nuclear fragmentation, formation of apoptotic bodies, and phagocytosis. ICE protein was negatively expressed in untreated control cells, while positive cells were expressed in apoptotic cells and apoptotic bodies. The number of positively expressed cells and staining intensity increased with prolonged drug action time, but some did not have withered. The ICE protein is also positively expressed in tumor cells in the state of death or in the pre-apoptotic state. The results showed that ICE gene may be involved in the regulation of apoptosis induced by cisplatin, and ICE protein may play a role in the early stage of apoptosis induced by cisplatin.
