胆碱酯酶活性的测定方法很多,目前国内多采用Hestrin的比色法和Ellman法测定。随着研究的深入,如有机磷毒剂中毒后残余酶活性的研究,对测定方法的灵敏度提出了更高的要求。国外,目前最灵敏的测定方法是放射化学测定法。一般应用~3H或~(14)C标记的乙酰胆碱为底物,在酶的作用下分解成标记的乙酸和非标记的胆碱。只要把标记乙酸和标记底物进行分离,即可测知酶的活性。可用于分离的方法很多,但以Johnson的用闪烁液直接提取并测定的方法最为简单。我们根据这一方法,并按照国内的条件,稍加改进,结果尚满意。
