细胞癌基因激活与细胞癌变有密切关系。由肿瘤组织提取DNA转染NIH/3T3细胞,得到恶性转化细胞,从中分离属于人高度重复序列Alu,其中含有致癌活性的DNA后段即转化基因。我们用上述方法发现人食管癌组织中有转化基因。从8例林县食管癌患者手术摘除的食管癌组织、北京胎儿食管上皮和10代以下的NIH/3T3细胞(日本 Dr Terada赠)提取DNA,用提取的DNA转染NIH/3T3细胞,5%新生牛血清筛选转化灶(Foci),以转化基因中人重复序列Alu为探针进行Southern法核酸杂交。
Cellular oncogene activation is closely related to cell carcinogenesis. NIH/3T3 cells were transfected with DNA extracted from tumor tissues to obtain malignant transformed cells. The human highly repeatable sequence Alu was isolated from the cells and contained transforming genes in the latter part of the carcinogenic DNA. We used the above method to discover transforming genes in human esophageal cancer tissues. From 8 cases of esophageal cancer tissue removed from patients with esophageal cancer in Linxian County, Beijing fetal esophageal epithelium and NIH/3T3 cells under 10 generations (gifted by Japan Dr Terada), DNA was extracted and NIH/3T3 cells were transfected with the extracted DNA, 5% Neonatal bovine serum was screened for foci, and the human repeat sequence Alu was used as a probe for Southern hybridization.
