[目的]从土壤链霉菌CaiF1发酵液中分离纯化抗菌物质,并研究该物质的活性。[方法]采用乙酸乙酯萃取,大孔吸附树脂吸附,硅胶层析和制备型高效液相色谱(HPLC)等分离技术纯化抗菌物质,以金黄色葡萄球菌、白粉病菌为指示菌进行活性研究。[结果]土壤链霉菌CaiF1发酵液中抗菌物质得到分离纯化;该物质在pH2.0-pH10.0、100℃和紫外照射24h条件下抑菌活性几乎不变,对白粉病的防治效果达69.7%。[结论]分离纯化的土壤链霉菌CaiF1抗菌物质具有很好的稳定性,为其结构鉴定和开发应用提供了理论依据。
[Objective] The research aimed to isolate and purify antibacterial substances from the fermentation broth of soil Streptomyces CaiF1 and study its activity. [Method] The antibacterial substances were purified by ethyl acetate extraction, macroporous adsorption resin adsorption, silica gel chromatography and preparative high performance liquid chromatography (HPLC). Staphylococcus aureus and powdery mildew were used as the indicator bacteria. [Result] The antibacterial substance in the fermentation broth of soil streptomyces CaiF1 was isolated and purified. The antibacterial activity of the material in the fermentation broth of CaiF1 at pH2.0-pH10.0, 100 ℃ and ultraviolet irradiation was almost unchanged, and the control effect on powdery mildew was 69.7 %. [Conclusion] The antibacterial substance of isolated and purified soil strain CaiF1 had good stability and provided theoretical basis for its structure identification and development and application.
