目的:探讨控制性促排卵(contral ovarian stimulation,COS)黄体期添加雌激素对小鼠子宫内膜白血病抑制因子(leukemia inhibitory factor,LIF)mRNA表达的影响。方法:将正常雌性昆明小鼠随机分为A组(单纯COS组)、B组(COS+黄体酮组)、C组(COS+黄体酮+雌激素组)和D组(自然周期,对照组),采用逆转录聚合酶链反应法检测LIF mRNA在各组小鼠妊娠第3~5天子宫内膜的表达情况。结果:4组小鼠妊娠不同时间子宫内膜LIF mRNA表达差异均有统计学意义(P<0.05);B,C,D组妊娠第4天时LIF mRNA表达水平均达峰值,而A组妊娠第3天其表达水平最高;妊娠小鼠子宫内膜种植窗高峰日LIF mRNA的表达,C,D组均明显高于B组(P<0.05),B组高于A组(P<0.05)。结论:添加雌激素较传统黄体支持更能改善小鼠子宫内膜LIF mRNA在小鼠子宫内膜的表达,增加子宫内膜容受性,纠正控制性促排卵对内膜种植窗前移的负面影响。
OBJECTIVE: To investigate the effect of estrogen on the expression of leukemia inhibitory factor (LIF) mRNA in the contral ovarian stimulation (COS) luteal phase. Methods: Normal female Kunming mice were randomly divided into group A (COS group), group B (COS + progesterone group), group C (COS + progesterone + estrogen group) and group D Reverse transcription polymerase chain reaction was used to detect the expression of LIF mRNA in the endometrium of the third to fifth days of pregnancy in each group. Results: The expression of LIF mRNA in endometrium of 4 groups at different times of pregnancy was significantly different (P <0.05). The levels of LIF mRNA reached the peak on the 4th day of pregnancy in groups B, C, D, The expression level of LIF mRNA in the endometrium of pregnant mice at the peak of implantation window was significantly higher than that of group B (P <0.05), and that of group B was higher than that of group A (P <0.05). Conclusion: The addition of estrogen can improve the expression of endometrial LIF mRNA in mouse endometrium, increase the endometrial receptivity, and correct the negative effect of controlled ovulation on endometrial plantar window shift influences.
