目的:探讨五田保肝液对大鼠酒精性肝纤维化的干预作用及对酒精性肝纤维化大鼠肝脏过氧化物酶体增殖物激活受体γ(PPARγ)表达的影响。方法:将100只Wistar大鼠随机分成6组:正常组10只、模型组、五田保肝液低、中、高剂量组、易善复组各18只,采用白酒辅以吡唑、玉米油的混合食料ig的方法建立大鼠酒精性肝纤维化模型,12周末处死大鼠,HE染色观察肝组织形态学改变;Masson染色观察肝纤维化情况;酶联免疫吸附法(ELISA)检测大鼠血清层黏连蛋白(LN),Ⅲ型前胶原(PⅢP)的水平变化;免疫组化法检测大鼠肝组织α-平滑肌肌动蛋白(α-SMA)蛋白的表达;实时定量RT-PCR法检测肝组织PPARγmRNA的表达。结果:与模型组相比,五田保肝液和易善复可减轻大鼠肝脏胶原纤维的增生,减少α-SMA蛋白的表达,使大鼠血清LN和PⅢP水平下降,肝组织PPARγmRNA的表达升高,且以五田保肝液中、高剂量效果更显著。PPARγmRNA的表达与α-SMA蛋白表达呈负相关。结论:五田保肝液对酒精性肝纤维化大鼠肝组织PPARγ表达具有促进作用,这可能是五田保肝液抑制大鼠酒精性肝纤维化发生发展的机制之一。
Objective: To investigate the effects of Wutian Liver Protection Liquid on alcoholic liver fibrosis in rats and its effect on the expression of liver peroxisome proliferator - activated receptor γ (PPARγ) in alcoholic liver fibrosis rats. Methods: 100 Wistar rats were randomly divided into 6 groups: normal group (n = 10), model group, Wuda liver preservation solution low, medium and high dose group, Oil mixture of food ig method to establish rat model of alcoholic liver fibrosis, rats were sacrificed at the end of 12 weeks, HE staining observed liver morphology; Masson staining observed liver fibrosis; enzyme-linked immunosorbent assay (ELISA) Serum laminin (LN) and type Ⅲ procollagen (PⅢP) were detected by immunohistochemistry. The expression of α-smooth muscle actin (α-SMA) Method to detect the expression of PPARγmRNA in liver tissue. Results: Compared with the model group, Wuda liver-preserving solution and Yishen Fu can reduce the proliferation of rat hepatic collagen fibers, reduce the expression of α-SMA protein, decrease the level of serum LN and PⅢP, and the expression of PPARγmRNA Increased, and Wu Tian liver solution, high-dose effect is more pronounced. PPARγmRNA expression and α-SMA protein expression was negatively correlated. CONCLUSION: Wuda Liver-preserving Liquid can promote the expression of PPARγ in the liver of alcoholic liver fibrosis rats, which may be one of the mechanisms that Wuda Liver-Protecting Liquid can inhibit the occurrence and development of alcoholic liver fibrosis in rats.
