目的通过正交实验优化胡黄连苷Ⅱ治疗大鼠脑缺血损伤的最佳剂量和时间窗。方法应用双侧颈总动脉结扎法建立大鼠前脑缺血模型,按照正交实验设计分组,经腹腔注射胡黄连苷Ⅱ干预治疗,酶联免疫吸附法检测脑组织中神经元特异性烯醇化酶(NSE)、神经胶质细胞标志蛋白(S100B)、髓鞘碱性蛋白(MBP)的含量,综合评价胡黄连苷Ⅱ治疗脑缺血损伤的效果。结果胡黄连苷Ⅱ治疗脑缺血损伤的最佳治疗时间窗和剂量为:①根据脑组织NSE含量分析为脑缺血1.5 h腹腔注射胡黄连苷Ⅱ10 mg·kg-1体重;②根据脑组织S100B含量分析为脑缺血1.5 h给予胡黄连苷Ⅱ20 mg·kg-1;③根据脑组织MBP含量分析为脑缺血1.5 h腹腔注射胡黄连苷Ⅱ10 mg·kg-1。结论根据治疗时间窗最大化和用药剂量最小化原则综合评价,胡黄连苷Ⅱ治疗脑缺血损伤的最佳治疗时间窗和剂量为脑缺血1.5 h腹腔注射10~20 mg·kg-1体重。
Objective To optimize the dose and time window of picroside Ⅱ for the treatment of cerebral ischemia in rats by orthogonal experiment. Methods The model of forebrain ischemia in rats was established by bilateral common carotid artery ligation. The rats were divided into groups according to the orthogonal experimental design. The rats were treated with picroside II by intraperitoneal injection. The neuron-specific enolase Enzyme (NSE), glial cell marker protein (S100B) and myelin basic protein (MBP), and to evaluate the effect of picroside II on cerebral ischemic injury. Results The optimal treatment time and dosage of picroside II for the treatment of cerebral ischemia were as follows: ① According to the analysis of NSE content in brain tissue, intraperitoneal injection of picrotoxin Ⅱ 10 mg · kg-1 body weight 1.5 h after cerebral ischemia; ② According to the brain tissue The content of S100B was given as picrotoxin Ⅱ20 mg · kg-1 1.5 hours after cerebral ischemia. ③ According to the MBP content of brain tissue, picrotoxinⅡ10 mg · kg-1 was intraperitoneally injected 1.5 hours after cerebral ischemia. Conclusion According to the principle of maximizing the treatment time window and minimizing the dosage of picroside Ⅱ, the best treatment time window and dose of picrotoxin Ⅱ for the treatment of cerebral ischemia were intraperitoneal injection of 10-20 mg · kg-1 body weight .
