目的:本实验主要研究脓毒症大鼠血清内毒素水平及脾脏TH1/TH2表达变化以及芪参活血颗粒对二者影响的研究。方法:采用盲肠结扎穿孔(CLP)的方法造成脓毒症模型,造模成功后随机分组,造模后3、24、72h腹腔静脉取血。采用基质显色法检测血清中内毒素含量,采用FACS流式细胞仪检测脾脏淋巴细胞TH1/TH2的表达。结果:结果表明脓毒症大鼠血清内毒素水平明显升高,芪参活血颗粒能明显降低血清内毒素水平,脓毒症大鼠早期TH1/TH2明显升高,后期明显下降,芪参活血颗粒能明显降低早期TH1/TH2的比值,升高晚期脾淋巴细胞TH1/TH2表达。使促炎与抗炎达到平衡。结论:芪参活血颗粒能减低脓毒症血清内毒素水平,调节TH1/TH2比值,是很好的免疫调节剂。
Objective: This study mainly focused on the study of serum endotoxin levels and splenic TH1/TH2 expression changes in sepsis rats and the effects of Qishen Huoxue Granules on them. Methods: The sepsis model was induced by cecal ligation and perforation (CLP). After successful modeling, the sepsis was randomly divided into groups. At 3, 24, and 72 hours after the model was established, blood was drawn from the abdominal veins. The serum endotoxin level was detected by matrix chromogenic assay. The expression of TH1/TH2 in splenic lymphocytes was detected by FACS flow cytometry. RESULTS: The results showed that serum endotoxin levels in sepsis rats were significantly increased, and Qishenhuoxue granules could significantly reduce serum endotoxin levels. The early TH1/TH2 levels in sepsis rats were significantly increased, and they were significantly decreased in later stages. Qishen Huoxue Granules It can significantly reduce the ratio of early TH1/TH2 and increase the expression of TH1/TH2 in late spleen lymphocytes. Make a balance between pro-inflammatory and anti-inflammatory. Conclusion: Qishenhuoxue granule can reduce the serum endotoxin level of sepsis and adjust the ratio of TH1/TH2. It is a good immunomodulator.
