目的:研究虎杖提取物对2型糖尿病大鼠血糖、血脂的影响。方法:通过高脂饮食加链脲佐菌素建立大鼠2型糖尿病模型,随机分为模型组、消渴降糖胶囊(75 mg/kg)阳性对照组及虎杖提取物高(888 mg/kg)、中(444mg/kg)、低(222 mg/kg)剂量组,另设正常对照组。连续灌胃给药4 w,每2 w检测1次空腹血糖,末次给药后检测血清中游离脂肪酸(FFA)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、糖化血清蛋白(GSP)、胰岛素(FINS)等指标,计算胰岛素抵抗指数(IRI)。结果:虎杖提取物各剂量均可显著降低2型糖尿病模型大鼠空腹血糖并改善其胰岛素抵抗,显著降低2型糖尿病模型大鼠血清FFA、TC、TG及LDL-C含量,提高血清HDL-C含量(P<0.05或P<0.01),并改善胰腺组织病理学改变;虎杖提取物中、低剂量可显著降低2型糖尿病大鼠GSP含量(P<0.05)。结论:虎杖提取物能改善2型糖尿病大鼠糖代谢与脂代谢和胰岛素抵抗情况,对治疗2型糖尿病具有一定疗效。
Objective: To study the effects of Polygonum Cuspidatum extract on blood glucose and blood lipid in type 2 diabetic rats. Methods: A rat model of type 2 diabetes mellitus was established by high fat diet and streptozotocin (STZ). The rats were randomly divided into model group, Xiaokejiangtang Capsule (75 mg / kg) positive control group and Polygonum cuspidatum extract high (888 mg / kg ), Medium (444mg / kg), low (222mg / kg) dose group, another set of normal control group. After continuous oral administration for 4 weeks, fasting blood glucose was measured every 2 weeks. After the last administration, serum FFA, TC, TG, HDL- C, LDL-C, GSP, FINS and so on. The insulin resistance index (IRI) was calculated. Results: Polygonum cuspidatum extract could significantly decrease fasting blood glucose (FBG) and insulin resistance in type 2 diabetic rats and significantly decrease serum levels of FFA, TC, TG and LDL-C in type 2 diabetic rats and increase serum HDL-C (P <0.05 or P <0.01), and improve the histopathological changes of pancreas. Low and medium doses of Polygonum Cuspidatum extract could significantly reduce GSP content in type 2 diabetic rats (P <0.05). Conclusion: Polygonum cuspidatum extract can improve glycometabolism, lipid metabolism and insulin resistance in type 2 diabetic rats and has some therapeutic effects on type 2 diabetes mellitus.
