为了探讨运动对长骨发育的作用机理,采用反转录聚合酶链反应(RT-PCR)技术,对100只生长期大鼠经3周、6周和9周的高、低负荷跑台训练后胫骨生长板中局部调节因子甲状旁腺素相关蛋白(PTHrP)的表达进行了观察。结果表明:3周的跑台运动没有显著改变生长板中PTHrP mRNA的表达水平,而6周和9周运动后PTHrP mRNA的表达水平显著高于对照组,低负荷运动组亦显著高于高负荷运动组(P<0.01或0.05);各组间对照组PTHrP mRNA的表达水平均未出现显著的增龄变化。结果提示:运动可以通过改变PTHrP的表达水平来调节生长板软骨细胞的活动,进而影响生长板的高度,调节长骨的纵向生长,但其具体的调节途径尚有待进一步研究探讨。
In order to explore the mechanism of exercise on long bone development, reverse transcription-polymerase chain reaction (RT-PCR) was used to train 100 rats in high and low-load treadmill after 3 weeks, 6 weeks and 9 weeks The expression of PTHrP, a local regulator of tibial growth plate, was observed. The results showed that: 3-week treadmill exercise did not significantly change the expression of PTHrP mRNA in the growth plate, while the expression of PTHrP mRNA at 6 weeks and 9 weeks after exercise was significantly higher than that in the control group, and also significantly higher than that in the low-load exercise group Exercise group (P <0.01 or 0.05). The expression of PTHrP mRNA in the control group did not show significant changes in aging. The results suggest that exercise can regulate the activity of growth plate chondrocytes by changing the expression level of PTHrP, thereby affecting the height of growth plate and regulating the longitudinal growth of long bones. However, the specific regulation of the pathway remains to be further studied.
