目的:观察氧比修饰低密度脂蛋白(OX-LDL)对血管内皮细胞的损伤作用。方法:在人脐静脉内皮细胞体外培养的基础上,通过采用3H胸腺嘧啶校苷(3H-TdR)掺入DNA技术和硫代巴比妥酸比色法,分别分析了OX-LDL对内皮细胞DNA合成及脂质过氧化物的代谢产物丙二醛(MDA)含量的影响。结果:当内皮细胞暴露于OX-LDL(50~200μg/ml)24小时后,其3H-TdR掺入量明显减少,但MDA含量显著增加。结论:OX-LDL能够明显抑制内皮细胞DNA合成,并显著增加内皮细胞的脂质过氧化物含量。
Objective: To observe the effect of oxygen-modified low-density lipoprotein (OX-LDL) on vascular endothelial cells. Methods: Human umbilical vein endothelial cells were cultured in vitro. 3H-TdR DNA incorporation and thiobarbituric acid colorimetric assay were used to analyze the effect of OX-LDL on endothelial cells DNA synthesis and the content of malondialdehyde (MDA), a metabolite of lipid peroxides. Results: When exposed to OX-LDL (50 ~ 200μg / ml) for 24 hours, the incorporation of 3H-TdR in endothelial cells decreased significantly, but MDA content increased significantly. Conclusion: OX-LDL can significantly inhibit endothelial cell DNA synthesis and significantly increase the lipid peroxidation of endothelial cells.
