目的建立一种快速溶剂萃取(ASE)-电雾式检测器(CAD)同时测定酸枣仁中酸枣仁皂苷A、酸枣仁皂苷B、斯皮诺素、白桦脂酸的方法。方法采用正交试验对快速溶剂萃取系统的提取条件进行优化,并用电雾式检测器对酸枣仁中目标成分进行同时检测。色谱柱为Thermo Syncronis C_(18)(100 mm×3 mm,3μm),流动相为乙腈-水,体积流量为0.5 mL/min,柱温为40℃,Corona Ultra电雾式检测器,雾化温度35℃。结果优化了ASE参数,极大地提高了提取效率,通过1次进样,同时测定酸枣仁中酸枣仁皂苷A、酸枣仁皂苷B、斯皮诺素、白桦脂酸的量,线性关系良好,相关系数为0.998 3~0.999 6,加样回收率在98.46%~102.02%。结论该方法快速、简便、准确可靠,可用于酸枣仁药材的质量控制。
Objective To establish a rapid solvent extraction (ASE) - electrospray ionization detector (CAD) for the simultaneous determination of jujuboside A, jujuboside B, spinosin and betulinic acid in jujube. Methods The orthogonal test was used to optimize the extraction conditions of the rapid solvent extraction system. The target components in the wild jujube were simultaneously detected by the electrospray ionization detector. The column was Thermo Syncronis C 18 (100 mm × 3 mm, 3 μm). The mobile phase consisted of acetonitrile-water, the volume flow rate was 0.5 mL / min and the column temperature was 40 ℃. The Corona Ultra electrospray detector Temperature 35 ℃. Results The parameters of ASE were optimized and the extraction efficiency was greatly improved. The simultaneous determination of the contents of jujuboside A, jujuboside B, spinosin and betulinic acid in the wild jujube kernels by one injection was good and the correlation was good The coefficient was 0.998 3 ~ 0.999 6, the recovery rate was 98.46% ~ 102.02%. Conclusion The method is rapid, simple, accurate and reliable and can be used for the quality control of Chinese medicinal materials.
