分别向未夹闭颈外动脉(A组)和夹闭颈外动脉(B组)的恒河猴(Macaque)的一侧颈总动脉注入甲基苯基四氢吡啶(MPTP),制造帕金森病(PD)灵长类动物模型。发现未夹闭组动物均需重复注射MPTP2~4次方可出现PD症状;而夹闭组动物均经1次手术获得成功。术后1个月脑核磁共振检查显示注射侧黑质区域密度减低、面积缩小。抗酪氨酸羟化酶免疫组化脑切片显示手术侧中脑多巴胺能神经元明显少于对侧。研究结果表明:夹闭颈外动脉向颈总动脉注入MPTP,是一种简便有效的建立灵长类偏侧帕金森病模型的方法。
Methyl-phenyl-tetrahydropyridine (MPTP) was injected into the common carotid artery of Macaque, both without external carotid artery (group A) and with external carotid artery (group B), respectively. Parkinson’s disease (PD) primate model. Found that uncovered group of animals were required repeated injection of MPTP2 ~ 4 times PD symptoms; and the clamp group of animals were successful in 1 operation. Brain MRI at 1 month after operation showed that the density of substantia nigra region on the injection side was reduced and the area was reduced. Anti-tyrosine hydroxylase immunohistochemical brain sections showed that dopaminergic neurons in the midbrain were significantly less than contralateral. The results show that: clamping the external carotid artery injection of common carotid artery MPTP, is a simple and effective method to establish primate hemiparkinsonism model.
