目的探讨细胞周期调控抑制因子p57kip2和视网膜母细胞瘤蛋白(Rb蛋白)在大肠腺癌发生发展中的作用。方法采用免疫组化技术SP法,检测p57kip2和Rb蛋白在37例大肠腺癌和26例正常大肠黏膜组织中的表达。结果p57kip2蛋白阳性表达率在大肠腺癌组织中为40.5%,显著低于正常大肠黏膜组织(χ2=5.039,P<0.05),并与大肠腺癌组织分化程度和淋巴结转移均有关(χ2=8.914,χ2=4.416,P<0.05);Rb蛋白阳性表达率在大肠腺癌组织中为51.4%,显著低于正常大肠黏膜组织(χ2=4.234,P<0.05),并与大肠腺癌淋巴结转移有关(χ2=14.596,P<0.05),但与组织分化程度无关(P>0.05);p57kip2蛋白阳性表达组Rb蛋白阳性表达率高于p57kip2蛋白阴性表达组,但两者之间呈正相关(r=0.369,P<0.05)。结论p57kip2和/或Rb蛋白的低表达可能在大肠癌发生中发挥重要作用并可能预测其恶性程度及转移,对治疗有指导作用;p57kip2和Rb蛋白在大肠癌细胞周期调控中可能存在着协同作用。
Objective To investigate the role of cell cycle regulatory inhibitor p57kip2 and retinoblastoma protein (Rb) in the development of colorectal adenocarcinoma. Methods Immunohistochemical SP method was used to detect the expression of p57kip2 and Rb in 37 cases of colorectal adenocarcinoma and 26 cases of normal colorectal mucosa. Results The positive rate of p57kip2 protein in colorectal adenocarcinoma was 40.5%, which was significantly lower than that in normal colorectal mucosa (χ2 = 5.039, P <0.05), and was correlated with the degree of differentiation and lymph node metastasis in colorectal adenocarcinoma (χ2 = 8.914 , χ2 = 4.416, P <0.05). The positive rate of Rb protein in colorectal adenocarcinoma was 51.4%, which was significantly lower than that in normal colorectal mucosa (χ2 = 4.234, P <0.05) (χ2 = 14.596, P <0.05), but not with the degree of differentiation (P> 0.05). The positive rate of Rb protein expression in p57kip2 positive group was higher than that in p57kip2 negative group (r = 0.369, P <0.05). Conclusions The low expression of p57kip2 and / or Rb protein may play an important role in the carcinogenesis of colorectal carcinoma and may predict its malignancy and metastasis, which may guide the therapy. There may be synergistic effects of p57kip2 and Rb proteins in the cell cycle regulation of colorectal cancer .
