目的:检测腺病毒载体在血管壁中的转基因效率。 方法:实验分两组:正常动脉组和损伤动脉组,应用携带Lacz 基因的重组腺病毒(Ad5CMV-lacz)转染正常和损伤后大鼠颈动脉壁细胞,并检测了损伤动脉Ad5CMV-lacz 的转染效率。Ad5CMV-Lacz 孵育时间为45 m in。 结果:未损伤动脉的Lacz(+ )细胞仅限于内皮细胞,而损伤动脉的新生内膜细胞、中膜细胞和部分外膜细胞均可被转染,其中新生内膜细胞的转染阳性率最高,为8.9% ~38.5% (总效率24.7% )。 结论:重组腺病毒载体可以高效转染血管壁平滑肌细胞。
Objective: To test the transgene efficiency of adenoviral vector in the vascular wall. Methods: The experiment was divided into two groups: normal artery group and injured artery group, normal and injured carotid artery wall cells were transfected with recombinant adenovirus carrying Lacz gene (Ad5CMV-lacz), and the effects of Ad5CMV-lacz Transfection efficiency. Ad5CMV-Lacz incubation time is 45 mins. RESULTS: Lacz (+) cells without damage to arteries were confined to endothelial cells, but neointima cells, mesangial cells and some adventitial cells that damaged the arteries were transfected. The positive rate of transfection of neointimal cells was the highest , 8.9% ~ 38.5% (total efficiency 24.7%). Conclusion: Recombinant adenovirus vector can efficiently transfect vascular smooth muscle cells.
