报告采用6种不同方法处理透射电镜样本44例(5例家兔肺、13例肝癌、20例人类胸腺、3例涎腺腺泡细胞癌、2例直肠类癌、1例胰腺癌)。结果:新鲜组织取材,2.5%戊二醛固定,置4℃冰箱3~5d,或置于磷酸缓冲液内4℃冰箱保存一月内的样本,其细胞超微结构基本不丢失;10%甲醛及时固定的组织其细胞膜性结构保存较好;甲醛固定后石蜡包埋的组织其细胞超微结构丢失较多,但部分膜性结构、分泌颗粒、张力原纤维仍可保存。因此,上述方法均可用于临床病理电镜诊断和鉴别诊断。2%多聚甲醛2.5%戊二醛固定的组织,尽管细胞超微结构可以保存,但细胞肿胀,胞突、微绒毛均消失。
Sixty-four transmission electron microscopy (5 rabbit lungs, 13 liver cancer, 20 human thymus, 3 salivary acinar cell carcinoma, 2 rectal carcinoid, and 1 pancreatic cancer) were treated by six different methods. Results: Fresh tissue samples were fixed with 2.5% glutaraldehyde, placed in refrigerator at 4 ℃ for 3 ~ 5d, or stored in phosphate buffer at 4 ℃ for one month. The ultrastructure of cells was almost not lost. 10% formaldehyde The fixed membranous tissue of the fixed tissue was well conserved. The paraffin embedded tissue after formaldehyde fixation lost more ultrastructure of the cell, but some membranous structures, secretory granules and tension fibril could still be preserved. Therefore, the above methods can be used for clinical pathological diagnosis and differential diagnosis. 2% paraformaldehyde 2.5% glutaraldehyde fixed tissue, although the cell ultrastructure can be preserved, but the cell swelling, cell processes, microvilli disappear.
